" Case 3. On the 6th of March, 1852, countenance increased, the pulse was no I had occasion to remove the finger of a longer perceptible, and the sounds of the robust, healthy-looking young man, in the heart could not be satisfactorily heard. Royal Infirmary. He was already under During the whole of this time, artificial the influence of chloroform when I entered the room, and as there had been some difficulty in producing complete anaesthesia, and the last of the chloroform in the bottle was already on the handkerchief, it was thought advisable by my friend in charge of its administration to keep up the inhalation, in order to produce a coma sufficiently profound to last until the completion of the operation. It was therefore left over his face, and I commenced and removed the finger, slowly disarticulating it from the metacarpal bone. I distinctly recollect hearing the man breathing quickly and shortly; and I also remember, that when just about to look for the vessels, my attention was attracted to his condition, by not any longer hearing the respiration. The handkerchief was still on his face. I took it off, and found, to my consternation, that the respiration had been diligently employed, but still the air appeared to enter the chest very imperfectly. I despaired. I felt certain that the man was dead, and that no human aid could restore him; and if it had not been that those standing near me urged me to persevere, I believe I should then have deserted the case as hopeless. Just at this time it occurred to me to put my finger in the mouth and draw forward the tongue, in order to secure there being no impediment to the air entering the lungs. Retaining it in this position, we again began the artificial respiration, and found that then the chest was fully expanded by each inflation. After keeping it up for a minute or two, the gentleman, who had all along kept his hand on the pulse, exclaimed, to our delight, that he could again feel it—' It was just like a slight flutter that reached the uppermost of his four

Binding of the volatile anesthetic chloroform to albumin demonstrated using tryptophan fluorescence quenching.
The site(s) of action of the volatile general anesthetics remain(s) controversial, but evidence in favor of specific protein targets is accumulating. The techniques to measure directly volatile anesthetic binding to proteins are still under development. Further experience with the intrinsic protein fluorescence quenching approach to monitor anesthetic-protein complexation is reported using chlo...
متن کاملAtomistic Study of Lipid Membranes Containing Chloroform: Looking for a Lipid-Mediated Mechanism of Anesthesia
The molecular mechanism of general anesthesia is still a controversial issue. Direct effect by linking of anesthetics to proteins and indirect action on the lipid membrane properties are the two hypotheses in conflict. Atomistic simulations of different lipid membranes subjected to the effect of small volatile organohalogen compounds are used to explore plausible lipid-mediated mechanisms. Simu...
متن کاملAntagonism of inhalant and volatile anesthetic enhancement of glycine receptor function.
Recent studies suggest that alcohols, volatile anesthetics, and inhaled drugs of abuse, which enhance gamma-aminobutyric acid, type A, and glycine receptor-activated ion channel function, may share common or overlapping molecular sites of action on these receptors. To investigate this possibility, these compounds were applied singly and in combination to wild-type glycine alpha(1) receptors exp...
متن کاملEffect of inhalation anesthetics on spin-labeled cholesterol containing DPPC vesicles.
We have investigated by means of electron spin resonance (ESR) spectroscopy the influence of three inhalation anesthetics, i.e. halothane, chloroform and diethyl ether, on the interfacial and hydrophobic region as well of 38 mol% cholesterol containing DPPC unilamellar vesicles. The study has been carried out in the temperature range 25-45 degrees C. The variation of the order parameter, S, vs ...
